Creatine Monohydrate
Creatine monohydrate is one of the most extensively researched dietary ingredients, particularly in the context of physical performance and body composition.
Because of its extensive scientific data, the International Society of Sports Nutrition (ISSN) has recognized creatine monohydrate as the most effective ergogenic nutritional supplement currently available. [1]
Research indicates that combining creatine monohydrate supplementation with resistance/weight training can lead to significant positive impact on performance output, lean muscle mass, strength gains, and recovery from exercise. [2, 3, 4]
Supplementing with creatine increases the amount of creatine stored in your muscle cells, known as "muscle creatine stores." This is crucial for sustaining energy levels during exercise, as creatine helps replenish adenosine triphosphate (ATP)—the body's primary energy source—especially during high-intensity activities where ATP is rapidly depleted.

Current evidence suggests that HMB accelerates the regenerative capacity of skeletal muscle after high-intensity or prolonged exercise, which contributes to enhanced athletic performance and muscle gains in active individuals. [6, 7, 8]
Moreover, when combined with creatine monohydrate, HMB may confer additive effects, leading to further improvements in body composition, physical performance, and recovery time [10].
Vitamin D3 (as Cholecalciferol)
Vitamin D is synthesized in the skin upon exposure to sunlight and is also found in various foods.
Research has shown that vitamin D can enhance the muscle protein synthesis rate, particularly when combined with other anabolic factors like insulin and HMB [12]. This is thought to be through the sensitization of the Akt/mTOR signaling pathway, a key regulator of protein synthesis in muscle cells.
The combined effect of vitamin D with HMB and insulin leads to further activation of protein synthesis, suggesting a synergistic role of vitamin D in enhancing muscle anabolism/growth.
Bioperine® Black Pepper Fruit Extract
The black pepper plant (Piper nigrum) is a flowering vine that bears fruits (peppercorns) containing roughly 2-7.4% piperine by mass. In comparison, BioPerine®, a standardized extract of black pepper, contains a minimum of 95% piperine by mass. Hence, as little as 5 mg BioPerine® provides as much piperine as 250 mg of most black pepper spices. (For reference, 1/4 teaspoon of ground black pepper weighs about 500 mg.)
Piperine, the major alkaloid found in black pepper, has been an emphasis of research in recent years due to its promising bioavailability-enhancing properties [13]. In other words, supplementing with BioPerine® helps increase the amount of active ingredient absorption in Transparent Labs Creatine HMB.
